Currently, the mechanisms of how cells sense their environment and then regulate their migration and patterning through cell-matrix interaction have not been understood. Here we show that these questions can be addressed by modeling the cell-substrate system as a pre-strained elastic disk attached to an elastic substrate via molecular bonds at the interface. Based on this model, we establish analytical and numerical solutions for the displacement and stress fields in both cell and substrate, as well as traction forces at the cell-substrate interface. We show that the cell traction generally increases with distance away from the cell center and that the traction-distance relationship changes from linear on soft substrates to exponential on stiff substrates. These results indicate that cell adhesion and migration behaviors can be regulated by cell shape and substrate stiffness. Our analysis also reveals that the cell traction increases linearly with substrate stiffness on soft substrates but then levels off to a constant value on stiff substrates. This biphasic behavior in the dependence of cell traction on substrate stiffness immediately sheds light on an existing debate on whether cells sense mechanical force or deformation when interacting with their surroundings. Finally, we show that the cell arrangement and orientation can be influence by the pattern geometry of substrate, and the effect of geometry can be further tuned by the substrate stiffness.

    季葆华,男,1971年生,北京理工大学教授,博士生导师。宇航学院力学系系主任。分别于1993和1998年在西安交通大学获学士学位和博士学位。1998年~2001年,在中国科学院力学研究所从事博士后研究。2001年~2004年,在德国马普金属研究所做博士后研究。长期从事复合材料和仿生材料及其相互作用的静态和动态力学行为、以及多尺度力学建模等方面的研究,取得了原创性的学术成果。目前主要从事仿生材料力学和细胞与分子力学方面的研究。在国内外重要学术刊物上发表或接收发表学术论文80余篇。目前兼任中国力学学会副秘书长,生物力学专业委员会委员、生物材料与仿生力学专业组副组长;《Molecular and Cellular Biomechanics》区域主编(Regional Editor),《Theo. Appl. Mech. Lett》、《Journal of Mechanics and Mems》等多种国际和国内期刊编委。2006年入选北京市科技新星计划;2009年入选教育部“新世纪优秀人才支持计划”;2009年年获中国力学学会青年科技奖;2010年获国家杰出青年科学基金。
